Objective: To study the methods of culture and amplification of human bone marrow stromal cells( BMSCs) in vitro, and to induce BMSCs differentiating into the neuron-like cells. Methods: The BMSCs were isolated primarily from bone marrow of adult, and purified by passage culture. The 5th passage of BMSCs were induced by all-trans-retinoic acid (RA) and immunocytochemical staining with anti-NSEMAP--tublin, and GFAP were also performed on cells after 7 days and 14 days induction. Results: Neuron-like cells appeared among BMSCs after BMSCs treated by RA for 24 hours. The Neuron-like cells induced for 7 days were showed -tublin positive, but NSEMAP-2 negative. However, Neuron-like cells induced for 14 days all were showed NSEJMAP-2 positive. And the -tublin positive rate was lower. The positive percentage of NSE protein expression was 48.5,0.2%. Conclusion: BMSCs can be amplificated and purified in short time, be induced into neuronal-like cell by RA..
